We’re pleased to share the SuFEx Handle Fragment Library, developed in collaboration between Melius Organics and OTAVA ltd.

This library comprises 74,543 fluorosulfate and sulfamoyl fluoride-containing fragments, designed to enable covalent modification of proteins via Sulfur(VI) Fluoride Exchange (SuFEx) chemistry. As a second-generation “click” reaction, SuFEx provides robust, selective reactivity under mild conditions, making it particularly suitable for fragment-based covalent ligand discovery.

🔹 Targets key nucleophilic residues such as lysine, tyrosine, and histidine
🔹 Facilitates covalent inhibitor development, enzyme profiling, and bioconjugation
🔹 Well-suited for applications in chemical biology, proteomics, and structure-based drug design
🔹 All fragments are feasible, synthetically accessible, and available for virtual or physical screening
🔹 Compounds are prepared on demand using in-stock building blocks with a typical turnaround of ~3 weeks

The structural diversity and electrophilic tuning of this library make it a valuable resource for exploring underexploited covalent binding strategies, especially beyond cysteine targeting.
